The chalcone was synthesized from cyclohexanone and an aromatic aldehyde by aldol condensation reaction, followed by cyclisation with thiourea results in hexahydro quinazoline. The third position is methylated by using methyl iodide, finally the mannich reaction was carried out with five different amines at second position of the hexahydro quinazoline to get five different S-Mannich bases. All the 5 synthesized new compounds were studied for their physiochemical characters. And their structures were confirmed by spectral studies (IR, NMR). These new derivatives were evaluated for analgesic and anti-inflammatory activity.

Two methods for simultaneous estimation of Montelukast sodium and Levocetrizine HCl in two component solid dosage forms have been developed. The methods employ the application of absorption corrected for interference, the first order derivative method. All these methods utilize methanol as a solvent. In absorption corrected for interference method montelukast sodium shows maximum absorption at a wavelength of 280.2 nm ,and levocetrizine HCl at 232.2 nm, where the linearity ranges for both the drugs were 5-30 μg/ml. At the wavelength of montelukast sodium levocetrizine HCl shows zero absorbance but at the wavelength of levocetrizine HCl, montelukast sodium shows some absorbance from this method the amount of montelukast sodium were determined at 280.2 nm, while at the wavelength of levocetrizine HCl we get the total absorption of both the drug, the amount of levocetrizine HCl were determined by applying the absorption corrected for interference method. In First order derivative method, the absorbance values at 270.8 nm and 224 nm of first derivative spectrum was used for the estimation of montelukast sodium and levocetrizine HCl respectively without mutual interference. This method obeyed Beers law in the concentration range of 10-60 μg / ml for montelukast sodium and 10-35 μg / ml for levocetrizine HCl, the results of analysis of both method have been validated statistically and recovery studies confirmed the accuracy of the proposed method.

The present paper deals with the development of RP-HPLC method for the determination of Nitazoxanide and Ofloxacin in bulk and in formulation using UV detector. Selected mobile phase was a combination of 0.005 M tetra butyl ammonium hydrogen sulphate and acetonitrile in the ratio of 45:55 and the wavelength selected was 240 nm. The flow rate 1.0 ml/min, and the injection volume was 20 μl. The sepration was performed at ambient temperature. Retention time of Nitazoxanide and Ofloxacin was found to be 6.051and 2.106. Linearity of the method was found to be 400 to 600 mcg/ml and 160 to 240 mcg/ml for Nitazoxanide and Ofloxacin respectively. Accuracy of the method was determined through recovery studies by adding known quantities of standard drug to the pre analyzed test solution and was found to be 99.08-99.81% and 100.18-100.65% for Nitazoxanide and Ofloxacin respectively.
